What is the mechanism of action of Premarin?

Premarin binds to nuclear receptor present on the estrogen responsive tissues. It  modulate the action of pituitary gland present in the brain, the production of Gonadotrophins, Luteinizing hormone (LH) and follicle stimulating hormone (FSH) through a negative feedback action. It decreases the elevated levels of these hormones in the postmenopausal women.

What is the correct Administration of Premarin?

Premarin comes in different doses strengths of 0.3, 0.45, 0.625, 0.9 and 1.25mg. These are tablet dosage form and should be taken orally.

  • In the case of menopausal, hypogonadism and postmenopausal osteoporosis. The prescribed dose of medicine is 0.3 mg/day for 25 days followed by 5 days off but this dose may be adjusted according to the requirement of patients..
  • For women patient who have ovarian failure, 1.25 mg  per day for 3 weeks followed by the one week off treatment.
  • In case if a patient has breast cancer, then take 10 mg Premarin per day for 3 months.
  • In case of osteoporosis take 0.3 mg Premarin per day.

What to do in case of missed dose of Premarin?

When a woman missed the dose of a single day, take the dose within 12 hours. If in a case if she misses two or more pills then she should take 2 pills for two consecutive days after the last day failure day and the rest should be continued.

What to do in case you take over-dose of Premarin?

In case of over dose of this drug, immediately go to the doctor or nearby hospital. The symptoms of overdose may be nausea, vomiting, stomach pain and breast tenderness.

What are the side-effects of Premarin?

The some side effects may be produced while using Premarin like pain in stomach and back, headache, depression, insomnia, dizziness, breast pain and tenderness, vaginal hemorrhage, nausea, weight gain.

What safety measures should be adopted while taking Premarin?

  • Check your breast from time to time for any unusual lumps or discharge.
  • If you are going to have surgery or are on complete bed rest then don't take Premarin.
  • Don't smoke or use tobacco because it may increase the risk of blood clot and stroke.
  • If the cardiovascular events are suspected then discontinue the drug as soon as possible.
  • It should not be taken with other oral contraceptive.
  • This medicine is always prescribed for only short duration.
